Miicrofluidic-based Oil-immersion Lenses for High Resolution Microscopy
نویسندگان
چکیده
Optical imaging systems with submicron resolution typically require the use of expensive, high numerical aperture (NA) microscope objective lenses. Here, we describe a novel Microfluidic-based Oil-Immersion Lens (μOIL) chip that can perform submicron and wide field of view imaging. Key element of the chip design is the use of high index of refraction, oil-immersion ball mini-lenses that have high NA (~0.8) and therefore very good optical resolution (0.6-0.7 μm). The μOIL chip can be part of a lab-on-chip system or it can used as an add-on module in low cost stereoscopes to enable high resolution imaging of biological samples in the lab, in the doctor’s office or in the field.

An Optofluidic Lens Array Microchip for High Resolution Stereo Microscopy
We report the development of an add-on, chip-based, optical module—termed the Microfluidic-based Oil-immersion Lenses (μOIL) chip—which transforms any stereo microscope into a high-resolution, large field of view imaging platform. The μOIL chip consists of an array of ball mini-lenses that are assembled onto a microfluidic silicon chip. The mini-lenses are made out of high refractive index mate...
متن کاملImmersion Meta-Lenses at Visible Wavelengths for Nanoscale Imaging.
Immersion objectives can focus light into a spot smaller than what is achievable in free space, thereby enhancing the spatial resolution for various applications such as microscopy, spectroscopy, and lithography. Despite the availability of advanced lens polishing techniques, hand-polishing is still required to manufacture the front lens of a high-end immersion objective, which poses major cons...
متن کاملTheory and Applications of Confocal Theta Microscopy
The outstanding role of confocal fluorescence microscopy in modern biological research results from its optical sectioning capability, which allows three-dimensional analysis of thick specimens (Shotton 1989). This property is due to the combination of point-like light sources and detectors, such that the illumination and detection volumes are restricted. Only their common volume is relevant to...
متن کاملReflected image of a strongly focused spot.
We describe the reflection of a strongly focused beam from an interface between two dielectric media. If the beam is incident from the optically denser medium, the image generated by the reflected light is strongly aberrated. This situation is encountered in high-resolution confocal microscopy and data sampling based on solid immersion lenses and oil immersion objectives. The origin of the obse...
متن کامل2,2'-thiodiethanol: a new water soluble mounting medium for high resolution optical microscopy.
The use of high numerical aperture immersion lenses in optical microscopy is compromised by spherical aberrations induced by the refractive index mismatch between the immersion system and the embedding medium of the sample. Especially when imaging >10 micro m deep into the specimen, the refractive index mismatch results in a noticeable loss of image brightness and resolution. A solution to this...
